Using a
proposal designed to evaluate CitroBio 5% against Listeria monocytogenes
on fish the test was performed as follows:
1. Six
haddock fillets were purchased
2. Prior to testing, three of the six were swabbed for L. monocytogenes.
3. The surface of the six fillets were inoculated with L. monocytogenes
(ATCC 19111)
and allowed a 1 hour contact time.
4. The surface of three of the fillets was flooded with a 1 ounce/gallon
solution of the CitroBio material.
5. After 5 minutes of contact time the six fillets were swabbed and
evaluated for the presence of L. monocytogenes.
Results
a. The
three initial swabs were negative for Listeria.
b. The
inoculated control samples were positive for L. monocytogenes.
c. The
treated samples were positive for L. monocytogenes.
Based
on these results it was decided that a zone of inhibition study be performed
to determine what level of CitroBio was effective. 1. MOX agar which
is used to propagate L. monocytogenes was selected as the test medium.
2. A 24 hour culture was used to inoculate the plate.
i. The
plates were allowed to dry.
4. A blank
was included to be sure that= there was no interference from other sources
5. The
test solution was prepared at 1, 2, 3, 4, and 5 ounces / gallon
6. Discs
that were 12 mm in diameter were used. They were saturated with the
different solutions and spotted on the plates. This was done in duplicate
on different plates.
7. The
plates were incubated for 24 hours and the size of the zones of inhibition
measured and recorded.
8. Listed
below are the results:
9. Based
ib the information generated, at the levels of 2oz to 5oz the activity
against L Monocytogenes was effective at these levels.
